Important compounds for click reactions of cy5 tetrazide

Click chemistry is a term coined in the last decade which is used to describe reactions that are high yielding, wide in scope create only by products that can be removed without chromatography, are stereospecific, simple to perform, and can be conducted in easily removable or benign solvents.

The use of cy5 tetrazide

Sulfo cy5 tetrazie is a tetrazine activated cy5 dye used for chemoselective labelling. The fluorescence used in the reaction is of a long wavelength and is not visible to the human eye. But due to the various advancements in technology it can be readily viewed by most imaging systems. The cy5 dyes are all water soluble labelling reagents. The cy5 conjugates are excited maximally at 648 nm and fluoresce maximally at665 nm. They are excited to about 98% of maximum with either a krypton or argon laser or to about 63% of the maximum with either a helium or a neon laser. The sulfopropyl group of our cy5 dyes provides increased fluorescence brightness improved photostability and lower nano specific binding, and all these functions are relly advantageous for improved imaging applications.

The use of cy5.5 NHS ester

This chemical is used widely by the chemist in dying procedures. It is a reactive dye for the labelling of amino groups in peptides proteins and oligonucleosides. Cy5.5 is a far-red emitting dye which is ideal for fluorescent measurements where background fluorescent is a concern. It is also suitable for an in vivo NIR imaging experiments. This reagent can replace normal NHS esters and is thus very important and beneficial for the reactions pertaining to click chemistry.  It is a dark blue powder with a molecular weight of 716.32. It is soluble in organic solvents like DMSO, DMF, dichloromethane and is insoluble in solvents like water. Thus this compound has several chemical properties and various other benefits, which if exploited properly and in an organised manner can be beneficial for click reactions.

The use of cy5 picolylazide

The cy5 picolylazide incorporates a copper chelating group to the azide structure and thus allows co diffusion during many click reactions.  It has many chemical properties which help not only in catalysing the click reactions but also help in the process of chelating the reagents. The kinetic enhancement made possible by the azide allows the chemists to perform site specific protein labelling on the surface of living cells. The detection sensitivity was also increased for the detection of alkyne modified proteins and RNA labelled by metabolic feeding. The copper catalysed alkyne cyclo addition has been used extensively for the conjugation immobilization and purification of biomolecules. Despite excellent reaction kinetics high specificity and bioorthoginality the azide has been used to a far lesser extent in cells and in vivo, because of toxicity cause by copper mediated generation of reactive oxygen species.

Thus the above mentioned chemical compounds are beneficial due to various chemical properties they possess and should thus be used accordingly.
